NeuroVue Dye Solid:

NeuroVue Red, Maroon and Jade are also available in solid form for researchers who would like to make their own dye-coated materials for application to tissue (e.g. use for coating glass/metal pins or pieces of hair to provide a more rigid support when smaller injections are required).

Images 1-3 show that the diffusion properties of NeuroVue (NV) Maroon and NV Red are well matched. Filter strips were inserted into the olivocochlear efferent bundle in the brainstem near the facial genu of a mouse and after 4 weeks at 36oC the cochlea was prepared as a flat mount and imaged. Both dyes exhibit comparable diffusion from the brainstem to the cochlea (Images 1 and 2) and the near identical overlap of both dyes (Image 3) suggests almost identical lateral and transcellular diffusion properties.

Images 4-6 show that the diffusion properties of NV Maroon and NV Orange are well matched. Filter strips were inserted into the olivocochlear efferent bundle in the brainstem near the facial genu of a mouse and after 2 weeks at 36oC the cochlea was prepared as a flat mount and imaged. Both dyes exhibit comparable diffusion from the brainstem to the cochlea (Images 4 and 5) and the near identical overlap of both dyes (Image 6) suggests nearly identical lateral and transcellular diffusion properties. The somewhat stronger labeling of NV Orange in more distant fibers indicates that this dye diffuses slightly more effectively.

Images 7-9 show the effects of tail length modification on transcellular diffusion. Filter strips co-coated with NV Red and a shorter tail analog of NV Maroon (PTIR284) were inserted into the olivocochlear efferent bundle in the brainstem near the facial genu of a mouse. After 2 weeks at 36 oC the cochlea was prepared as flat mount and imaged. As before for this diffusion time, NV Red labeled fibers are found around inner hair cells and some fibers extent to outer hair cells (Image 7). In contrast the green false colored shorter tail analog of NV Maroon (PTIR284) shows dramatic transcellular diffusion of almost all cells in the cochlea (green circles, black holes are nuclei, Image 8). The superposition of both dyes (Image 9) shows how further the shorter tail NV Maroon analog (green) has spread out compared to the longer tail NV Red (red). These results show that transcellular diffusion properties are strongly dependent on alkyl ‘tail’ number and length and suggest that transcellular diffusion maybe helpful in studying contacts in difficult to access tissue.
